Three novel mixed ligand Zn(II) complexes {complex 1 = [Zn(L1)(2)(bpy)], complex 2 = [Zn(L2)(2)(bPY)], complex 3 = [Zn(L2)(2)(phen)], where bpy = 2,2′-bipyridine and phen = 1,10-phenanthroline} of 4-(4-chlorobenzoyl)-2-(3-chlorophenyl)-5-methyl-2,4-dihydro-3H-pyrazol-3-one (HL1) and 4-(4-chlorobenzoyl)-5-methyl-2-(p-tolyl)-2,4-dihydro-3H-pyrazol-3-one (HL2) were synthesized and characterized by H-1 and C-13 NMR, FTIR, TG-DTA, UV-Vis, elemental analysis, molar conductance and single crystal X-ray structure determination. Based on analytical and spectroscopic techniques, all the three Zn(II) complexes were found to have a distorted octahedral geometry. The ligands and the complexes were screened for antimalarial activity against plasmodium falciparum. The zinc(II) complexes showed greater antimalarial activity than their parent ligands. A series of pyrazole ligands named as 1,4-bis(3,5-dimethylpyrazol-1-yl)butane (bbd), 1,3-bis(3,5-dimethylpyrazol-1-yl)propane (bdpp), and 1,3-bis(3,5-dimethylpyrazol-1-yl)propane-2-ol (bdpo) along with their transition metal complexes including [Cu-2(mu(3)-SCN)(2)(mu-bbd)](n), [Cu-2(mu(3)-SCN)(2)(mu-bdpp)](n), and [Cu2Hg(mu(2)-SCN)(2)(bdpo)(2)(CH3COO)(2)](n)were synthesized and characterized using FT-IR and Uv-vis spectroscopies, as well as thermal gravimetric analysis. The prepared compounds were investigated as corrosion inhibitors to protect mild steel in 0.5 M H(2)SO(4)solution. Corrosion tests were performed by electrochemical impedance spectroscopy and potentiodynamic polarization measurements. The pyrazole ligands exhibited inhibition efficiencies in a range of 38-78%, which introduced them as moderate inhibitors. On the other hand, the pyrazole complexes showed inhibition efficiencies higher than 85%, demonstrating their high potentials to mitigate the steel corrosion. In addition, the bimetallic complex resulted in higher corrosion resistance than the two monometallic complexes. This was evidence of a critical role of the coordinated metal of the complex in forming an adhesive protection film on the steel surface.

The Pd(ii)-catalyzed activation of Csp(2)-H bond and double alkyne annulation which proceeds via allylic isomerization is reported for the first time. This reaction of antipyrines with alkynes provides an efficient synthetic route for the biologically important spiro-cyclopentadiene pyrazolones. In the presence of Lawesson’s reagent, this Pd(ii)-catalyzed annulation reaction affords another spiro-cyclopentadiene pyrazolone which displays very good fluorescence properties.

An enantioselective Mannich reaction between 3-fluorooxindoles and pyrazolinone ketimines has been developed for the construction of amino-pyrazolone-oxindoles containing stereogenic C-F units. Based on this new protocol that allows for the generation of two adjacent tetrasubstituted stereocenters, a variety of structurally diverse fluorinated amino-pyrazolone-oxindoles were obtained in good to excellent yields with excellent diastereoselectivities and enantioselectivities (up to 98% yield, >20 : 1 dr and >99% ee). What’s more, good yield and high stereoselectivities were obtained in the gram-scale reaction.

Purpose: Regorafenib is an oral multikinase inhibitor approved for the therapy of previously treated metastatic colorectal carcinoma (mCRC). The aim of the present study was to analyze the outcomes of treatment with regorafenib in real-world clinical practice based on data from a national registry. Methods: The CORECT registry, the Czech non-interventional database of patients with mCRC treated with targeted agents, searched for patients with metastatic CRC treated with regorafenib. In total, 555 evaluable patients were identified. Results: The median age at diagnosis was 61.7 years. All patients had disease progression on or after previous systemic treatment. Most patients were treated with an initial dose of 160 mg daily (n = 463; 83.6%). The median duration of treatment was 2.7 months (range 0.023.4 months). By the data cut-off date, 472 patients (85%) had completed treatment with regorafenib and were evaluable for treatment response evaluation. Partial response was reported in 13 patients (2.8%) and disease stabilization in 130 patients (27.5%). Median progression-free survival (PFS) and overall survival (OS) were 3.5 months (95% confidence interval [CI] 3.2-3.7 months) and 9.3 months (95% CI 8.3-10.3 months), respectively. The 6-month OS rate was 67.7% (95% CI 63.4-72.1%). Multivariable analysis showed that female gender, longer interval from diagnosis of metastatic disease, MO stage at diagnosis, and Eastern Cooperative Oncology Group performance status (ECOG PS) 0 were associated with longer PFS, while higher body-mass index (BMI), longer interval from diagnosis of metastatic disease, and ECOG PS of 0 were associated with longer OS. Conclusion: OS of patients treated with regorafenib in the real-world clinical practice in this cohort exceeded that reported in randomized trials. Regorafenib is a safe and active treatment option for a subgroup of patients with mCRC who are progressing after other systemic therapies and maintain good performance status.

A cobalt-catalyzed regio- and enantioselective gamma-amination of beta,gamma-unsaturated N-acylpyrazoles that delivers the corresponding gamma-amination products in good regio- and enantioselectivity has been established. Moreover, the nitrogen-containing compounds could be easily synthesized. DFT calculations have been provided to explain regio- and enantioselectivity for this gamma-amination. The chiral gamma-amination products were readily converted into the chiral gamma-amino acid derivatives.

Synthesis of the multidentate coordinated chelate N3C-H-2, composed of a linked functional pyridyl pyrazole fragment plus a peripheral phenyl and pyridyl unit, was obtained using a multistep protocol. Preparation of Ir(III) metal complexes bearing a N3C chelate in the tridentate (kappa(3)), tetradentate (kappa(4)), and pentadentate (kappa(5)) modes was executed en route from two nonemissive dimer intermediates [Ir(kappa(3)-N3CH)Cl-2](2) (1) and [Ir(kappa(4)-N3C)Cl](2) (2). Next, a series of mononuclear Ir(III) complexes with the formulas [Ir(kappa(4)-N3C)Cl(py)] (3), [Ir(kappa(4)-N3C)Cl(dmap)] (4), [Ir(kappa(4)-N3C)-Cl(mpzH)] (5), and [Ir(kappa(4)-N3C)Cl(dmpzH)] (6), as well as diiridium complexes [Ir-2(kappa(5)-N3C)-(mpz)(2)(CO)(H)(2)] (7) and [Ir-2(kappa(5)-N3C)-(dmpz)(2)(CO)(H)(2)] (8), were obtained upon treatment of dimer 2 with pyridine (py), 4-dimethylaminopyridine (dmap), 4-methylpyrazole (mpzH), and 3,5-dimethylpyrazole (dmpzH), respectively. These Ir(III) metal complexes were identified using spectroscopic methods and by X-ray crystallographic analysis of representative derivatives 3, 5, and 7. Their photophysical and electrochemical properties were investigated and confirmed by the theoretical simulations. Notably, green-emitting organic light-emitting diode (OLED) on the basis of Ir(III) complex 7 gives a maximum external quantum efficiency up to 25.1%. This result sheds light on the enormous potential of this tetradentate coordinated chelate in the development of highly efficient iridium complexes for OLED applications.

Importance: treatment of dementia in individuals with comorbidities is complex, leading to potentially inappropriate prescribing (PIP). The impact of PIP in this population is unknown. Objective: to estimate the rate of PIP and its effect on adverse health outcomes (AHO). Design: retrospective cohort. Setting: primary care electronic health records linked to hospital discharge data from England. Subjects: 11,175 individuals with dementia aged over 65 years in 2016 and 43,463 age- and sex-matched controls. Methods: Screening Tool of Older Persons’ Prescriptions V2 defined PIP. Logistic regression tested associations with comorbidities at baseline, and survival analyses risk of incident AHO, adjusted for age, gender, deprivation and 14 comorbidities. Results: the dementia group had increased risk of PIP (73% prevalence; odds ratio [OR]: 1.92; confidence interval [CI]: 83-103%; P < 0.01) after adjusting for comorbidities. Most frequent PIP criteria were related to anti-cholinergic drugs and therapeutic duplication. Risk of PIP was higher in patients also diagnosed with coronary-heart disease (odds OR: 2.17; CI: 1.91-2.46; P < 0.01), severe mental illness (OR: 2.09; CI: 1.62-2.70; P < 0.01); and depression (OR: 1.81; CI: 1.62-2.01; P < 0.01). Two new Ni(II) coordination complexes of pyrazole and acetato ligands, involving nitrate and chloride counter anions, viz. [Ni(Hdmpz)(2)(CH3COO)(H2O)(3)]NO3 center dot 2H(2)O (1) and [Ni(Hdmpz)(2)(CH3COO)(H2O)(3)]Cl (2) (where Hdmpz = 3,5-dimethylpyrazole), have been synthesized in purely aqueous medium at room temperature. The complexes have been characterized by elemental analysis, FT-IR, electronic spectroscopy, PXRD, TGA and single crystal X-ray diffraction. The H-bonds established between the isostructural cationic [Ni(Hdmpz)(2)(CH3COO)(H2O)(3)](+) moieties of both complexes can be classified as anti-electrostatic H-bonds (AEHBs) and these have been studied using DFT calculations. Remarkably, in one case these AEHBs exhibit favorable interaction energies, thus indicating that the attractive nature of the H-bonds is able to compensate the pure electrostatic repulsion. The analyses of the cell viability results for the DL cell line show a significant concentration dependent decrease by compounds 1 and 2 with a negligible cytotoxic effect observed against normal cells (PBMC) in comparison to the cancer cells (DL). The cytotoxic activity of these compounds in DL cells occurs via apoptosis. Molecular docking and SAR analysis reveal that the compounds possess high binding affinities against most common cancer target proteins. (C) 2019 Elsevier Ltd. All rights reserved.
